摘要 目的观察超声波对前脂肪细胞体外培养与增殖的影响,论证超声辅助吸脂术后前脂肪细胞作为脂肪组织工程种子细胞的可行性。方法将经过超声波作用后(8min,3w/cm2)的猪前脂肪细胞进行体外培养,并每隔2d测定其细胞数量及脂质含量,观察其细胞增殖速率及脂质含量的变化,与正常对照组相比较。结果实验组细胞贴壁时间为1~2d,细胞倍增时间为72h,脂滴出现时间为培养后6d,脂质测定时其吸光度峰值为0.32,而正常对照组各项对应指标分别为6~24h、36h、4d,0.68;说明经超声波处理的猪前脂肪细胞体外培养有延迟贴壁、细胞增殖速率减缓、脂质含量减少等改变。结论超声波对前脂肪细胞的增殖分化有明显的抑制作用,经超声波处理的前脂肪细胞不宜用作种子细胞。 Objective To observe the effects of ultrasound on the proliferation and differentiation of porcine preadipocyte cultured in vitro and to investigate the feasibility of preadipocyte transplantation after ultrasound assisted liposuction. Methods The porcine preadipocyte were treated with ultrasound (8 min, 3 W/cm^2), purified and cultured in vitro. The rate of proliferation and the content of lipid were studied and compared with the control groups. Results The time of adhesion in experimental groups was 1 to 2 days, and the time of double-proliferation was about 72 h, the time when lipid could be detected was 6 days after culture, and the peak value of absorbance was 0.32. The values of those in control groups were 6 to 24 h, 36 h, 4 days, and 0.68, respectively. It was indicated that the number of adhesive cells in experimental groups were decreased, the time of adhesion delayed, the rate of proliferation became slow and the contents of lipid lowere also as compared with the control groups. Conclusion The proliferation and differentiation of porcine preadipocyte could be prohibited by ultrasound significantly and indicated that the preadipocytes treated with ultrasound could not be used as seed-cells.
