p21基因转染人胃癌细胞对化疗药物敏感性的影响 被引量:9

摘要 目的:探讨p21基因转染人胃癌细胞系(BGC)对化疗药物的敏感性。方法:应用MTT摄入方法测定5-氟尿嘧啶、阿霉素、长春新碱、环磷酰胺、平阳霉素和替尼泊甙对p21基因转染人胃癌细胞增殖抑制作用,并进行72 h观察。结果:p21基因转染人胃癌细胞对5-氟尿嘧啶最敏感,72 h抑制率为77.9%;其次为阿霉素、长春新碱、环磷酰胺、平阳霉素和替尼泊甙,其抑制率分别为62.7%、61.1%、59.3%、58.2%及45.9%。其敏感性与对照组相比差异有显著性(P<0.05),随时间延长而逐渐增加。结论:用MTT法检测p21基因转染人胃癌细胞对化疗药物敏感性的实验,即可以针对性指导临床对肿瘤敏感性化疗药物的选用,又可避免盲目用药而产生对机体免疫细胞的毒性作用。p21基因可提高对5-氟尿嘧啶的敏感性。 Objective To study the sensitivity of p21-transfected human gastri, cancer cell line (BGC) to chemotherapeutic drugs. Methods MTT assay was used to test the inhibitory effects of Fluorouracilum (5-Fu), Adriamycin (ADM), Vincristin (VCR), Cytoxan (CTX), Pingyanmycin (PYM) and Teniposide (VM-26) on p21-transfected human gastric cancer cells. The sensitivity of p21-transfected human gastric cancer cells to chemotherapeutic drugs was observed for 72 h. Results p21-transfected gastric cancer cells were the most sensitive to 5-Fu, then ADM, VCR, CTX, PYM and VM-26. The inhibitory rate was 77.9%, 62.7%, 61.1%, 59.3%, 58.2% and 45.9%, respectively. The difference of chemosensitivity compared with control was significant (P〈0.05). The sensitivity increased with prolongation of exposure time. Conclusion MTT assay is used to test the sensitivity of the gastric cancer cells transfected with p21 gene to chemotherapeutic drugs, Pretreatment determination of chemosensitivity can not only screen out sensitive drugs for individual tumor, but also avoid toxicity of ineffectual drugs to immune cells, p21 gene may increase the sensitivity of cancer cells to 5-Fu.
