转座元件介导的苯胺代谢基因簇的筛选和鉴定 被引量:2

摘要 利用苯胺不完全降解时中间代谢产物,如邻苯二酚等的显色反应,建立了简便快速的苯胺代谢途径相关基因的功能筛选方法.从苯胺降解菌株AD9的基因组文库中筛选得到3个阳性重组子pDA1,pDB2和pDB11.AD和C23O酶活测定结果表明,pDA1和pDB11分别含有功能完整的苯胺双加氧酶或邻苯二酚双加氧酶编码基因.经核苷酸序列分析和拼接获得一个大小为24.7kb,含25个可读框的苯胺代谢基因簇,其中含有17个基因与苯胺代谢有关,包括调控基因tadR,苯胺双加氧酶基因簇tadQTA1A2B,邻苯二酚间位裂解途径酶系基因簇tadD1C1D2C2EFGIJKL.苯胺代谢基因簇两侧含有转座酶和IS1071序列.
