摘要
目的:建立人胰岛素的转基因小鼠模型。方法:通过原核显微注射法,把外源基因pEF1α-m INS注射入FVB种小鼠受精卵,胚胎移植给同期发情的假孕受体母鼠获得子代小鼠。结果:移植注射胚胎204枚给38只假孕小鼠共出生108只后代鼠,经PCR和Southern b lot检测到5只阳性小鼠。结论:通过显微注射法使外源基因pEF1-αm INS在小鼠基因组中得到整合,建立了人胰岛素的转基因小鼠模型。
Objective: To construct transgenic mice model expressing human insulin. Methods: pEF1α-mINS fragments were microinjected into fertilized mice eggs. Those eggs were transplanted into the oviducts of the pseudopregnant mice. Results:Transgenic FVB mice were obtained by microinjection. PCR and Southern blot analysis showed 5 of 108 mice were transgenic. Conclusion:pEF1α-mINS fragment can integrate into mice genomic DNA. An animal model of transgenic mice expressing human insulin can be successfully established.
