摘要
目的:检测不同浓度与作用时间的钙离子(Ca2+)对HeLa细胞增殖与宫颈癌组织3H-TdR掺入的影响,同时检测维生素D3(VD3)的协同作用与维拉帕米(VP)的拮抗作用,探讨Ca2+抑制宫颈癌细胞与组织增殖的可能作用机制。方法:应用MTT法检测HeLa细胞在各作用因素存在时的增殖水平;应用同位素放射自显影检测新鲜人宫颈癌组织在各刺激因素存在时的3H-TdR掺入水平。结果:Ca2+具有抑制HeLa细胞增殖和宫颈癌组织3H-TdR掺入作用,VD3可明显协同增强Ca2+的抑制作用,VP可明显拮抗Ca2+的作用、部分拮抗VD3的协同作用。结论:Ca2+具有剂量与时间依赖性抑制宫颈癌细胞与组织增殖的作用,其作用机制与促进细胞外钙内流相关;VD3的协同抑制作用不仅与促进细胞外钙内流相关,还存在其它机制有待探讨。
Objective: To investigate the effects of different concentrations of calcium on cell proliferation in human cervical cancer cells and tissues at different times, to investigate the synergism of vitamin D3 (VD3 ) and the antagonism of verapamil (VP), and to explore the possible mechanisms. Methods:The level of HeLa cell proliferation in the presence of various factors was determined by MTT assay. The level of ^3H-TdR incorporation into the fresh human cervical cancer tissues in the presence of various stimulators was evaluated by autoradiography (ARG). Results:The results showed that calcium inhibited the proliferation of HeLa cell and the incorporation of 3 H-TdR into the cervical cancer tissues, and VD3 synergized the inhibitory effect of calcium. Calcium antagonist VP remarkably antagonized calcium-induced inhibitory effect and partly antagonized the synergism of VD3. Conclusion:Calcium could inhibit the proliferation of cervical cancer tissues and cells in a manner of dose- and time-dependent, the mechanisms of which were related with promoting the influx of extracellular calcium into cells. The synergism of VD3 is relevant not only to its facilitation of the influx of extracellular calcium into cell but also to other mechanisms, which need to be studied further.
