氯沙坦对抑制一氧化氮合酶Heymann肾炎大鼠的肾脏保护作用 被引量:2

摘要 目的:观察氯沙坦对抑制一氧化氮合酶(NOS)的Heym ann肾炎大鼠的肾脏保护作用。方法:将NOS抑制剂NW-硝基-L-精氨酸(L-NNA)和氯沙坦应用于Heym ann肾炎大鼠,21 d后测定血压,尿蛋白,计算肌酐清除率(Ccr),观察肾脏形态学改变,免疫组化法检测转化生长因子β1(TGF-β1)的表达。结果:抑制NOS后,Heym ann肾炎大鼠血压升高,尿蛋白增加,Ccr降低,形态学检查可见肾小管间质炎性细胞浸润,部分肾小球缺血,TGF-β1表达增加(P<0.01),加用氯沙坦后均得到改善。结论:抑制NOS加重Heym ann肾炎大鼠肾脏损害,同时应用氯沙坦具有肾脏保护作用。 Objective: To observe the protective effects of losartan on kidneys in rats with Heymann nephritis aggravated by nitric oxide synthase (NOS) inhibitor, Nw-nitro-L-arginine (L-NNA), Methods:L-NNA was administered to rats with Heymann nephritis, Tail blood pressure and urinary protein were measured. Creatinine clear rate (Ccr) was calculated, Renal morphological changes were evaluated at the 21 th day, The expression of transforming growth factor β1 (TGFβ1) was determined immunohistochemically. Results:L-NNA induced hypertension and the proteinuria, reduced Ccr, and led to peritubular inflammatory cell infiltration and some glomerular ischemic changes. Immunohistochemical staining showed that TGFβ1 deposition increased in L-NNA group ( P 〈0.01 ). Losartan prevented these damages. Conclusion:Inhibiting NOS induces renal damage in rats with Heymann nephritis. Losartan could prevente L-NNA-indueed renal injury in Heymann nephritis.
