摘要
本文用离子对高效液相色谱法(HPLC)测定血浆中的头孢克罗(Cefaclor)浓度。血浆样品用1%磷酸甲醇溶液提取。色谱条件为:紫外检测波长λ265nm;分析柱UltrasphereTM“IP”250mm×4.6mm;流动相甲醇:四氢呋喃:水(每升水中含1.0g庚磺酸钠,11g三乙胺,20.5g磷酸)。本法最小检测限50ng/ml。本法对10名受试者口服250mg头孢克罗后进行药动学和相对生物利用度研究。头孢克罗为一房室模型,其T1/2(ke)0.6h,Tmax0.4~0.7h,Cmax4.92~6.50μg/ml。颗粒剂的相对生物利用度为1.07±0.13。
efaclor is a new cephalosporin antibiotic for oral use. Cefaclor in plasma was deterrnined hy IP-HPLC. Theplasma sample was extracted with a mixture solution of phosphoric acidand methanol. The mobile phase wasmethanol :tetrahydrofuran :water (contain 1.0g heptanesulfonate sodium , 11.0g triethylamine and 20. sg phospho-ric in 1. 0L water). Analrtical column was Ultrasphere TM'IP''(250mm × 4. 6mm ID). The detection wavelengthwas 265nm. The minimun detective concentration was 50 ng/ml. Using this method, pharmacokinetics andbioavailaility of cefaclor in ten male healthy volunteers after oral administration of 250mg dosage was studied. Theplasma-time concentration curve of volunteers was one-compartment model. The half-life time of elimination was0. 6h,Tmax 0. 4h ̄ 0. 7h and Cmax .4. 92and 6. 50μg/ml. The relative bioavailability of cefaclor granule was 1. 07 ±0. 13.
