摘要
目的:检测大鼠实验性骨关节炎(OA)关节软骨中的p53蛋白和mRNA在不同时期的表达,明确其在OA中的作用。方法:采用经典的Hu lth法构建大鼠膝关节实验性OA的动物模型。免疫组化方法检测p53蛋白的表达。RT-PCR技术检测p53 mRNA的表达。结果:OA关节软骨逐渐变为淡黄白色,粗糙。HE染色见OA软骨细胞变得稀疏,集聚成簇。免疫组化表明OA模型侧关节软骨p53蛋白高表达。RT-PCR表明OA侧p53 mRNA高表达,而且随时间增加而表达持续上调。结论:本研究表明在OA关节软骨的p53蛋白和mRNA的高表达与OA进程密切相关,可能是OA的发病机制之一。
Objective : To detect the expression of p53 protein and mRNA in articular cartilage of rats with experimental osteoarthritis (OA) in different periods and to determine their functions in osteoarthritis. Methods:The animal models of rat with knee joint experimental osteoarthritis were established with Hulth method. The expression of p53 protein was detected by using immunohistoehemical method. The expression of p53 mRNA was detected by using RT-PCR techniques. Results :The cartilages of 0steoarthritis models were slightly white-yellow and rough. HE staining showed that the chondrocytes were sparse and some assembled into clusters. Immunohistoehemical results showed the expression of p53 protein increased in osteoarthritis models. The RT-PCR showed the expression of p53 mRNA increased. Moreover, the increase was directly correlated with time. Conclusion :The increase of p53 protein and mRNA expression in articular cartilage of osteoarthritis is closely correlated with the disease process, which may be the pathogenesis of osteoarthritis.
