摘要
目的:探讨犬胆管损伤后增殖平滑肌细胞受辐射后细胞增殖抑制和凋亡发生的关系。方法:在犬胆管内分别植入103Pd放射性支架和普通胆管支架,取出胆管标本采用ABC法进行增殖细胞核抗原(PCNA)免疫组化染色和平滑肌细胞凋亡染色。结果:103Pd支架组犬胆管组织平滑肌细胞PCNA蛋白阳性表达率为33.33%,而普通支架组犬胆管组织平滑肌细胞PCNA蛋白阳性表达率为83.33%,103Pd支架组PCNA蛋白阳性表达率明显低于普通支架组(P<0.01)。而103Pd放射性支架组犬胆管增殖平滑肌凋亡细胞数明显大于普通支架组犬胆管增殖平滑肌细胞数(P<0.05),而且103Pd放射性支架组犬胆管腔无明显狭窄,而普通支架组犬胆管腔有明显狭窄。结论:辐射可以抑制平滑肌细胞增殖,同时可诱导犬胆管平滑肌细胞凋亡,抑制犬胆管再狭窄。
Objective: To study the relationship between the inhibited proliferation and induced apoptosis of biliary smooth muscle cells caused by radiation in dogs with biliary injury, nethods:^103Pd-containing stent and normal stent were placed in the biliary tract of dogs with biliary injury. The expression of proliferating cell nuclear antigen (PCNA) was detected by ABC immunohistochemical staining in the biliary smooth muscle cells. Results:There were significant differences in the positive expression rate of PCNA in biliary smooth muscle cells (33.33% vs. 83.33%, P 〈 0.01 ) and in the number of apoptosis cells[87.9 ±7.96 vs. 5.6 ± 7.96, P 〈 0.05 ] between ^103Pd-containing stent group and normal stent group. The stricture of the biliary tract was more obvious in normal stent group than in ^103Pd-containing stent group. Conclusion: Radiation may prevent the stricture of biliary tract by inhibiting the proliferation and inducing the apoptosis of biliary smooth muscle cells.
