摘要
目的:观察苹果酸、γ-氨基丁酸(GABA)对Hela细胞端粒酶活性的影响。方法:苹果酸、γ-氨基丁酸作用于Hela细胞不同时间后,用TRAP-银染半定量法检测端粒酶活性的变化。结果:1.将不同浓度的苹果酸作用于Hela细胞48 h后,端粒酶活性降低,以10μmol/L浓度时作用活性最低。10μmol/L苹果酸作用于Hela细胞不同时间,端粒酶活性于作用8 h时就有所下降,当作用48 h时与对照组相比下降最多达到11%。2.将不同浓度的GABA作用于Hela细胞48 h后,端粒酶活性降低,以1 mmol/L浓度时作用活性最低。1 mmol/L GABA作用于Hela细胞不同时间,结果显示端粒酶活性于作用8 h时就有所下降,当作用48 h时与对照组相比几乎下降最多达到13%。结论:苹果酸,GABA对Hela细胞端粒酶活性起下调作用。
Objective:To observe effect of malate and gamma-aminobutyric(GABA) on telomerase activity of Hela cells, Methods:Hela cells were treated with various concentration of malate and GABA respectively for different time and then the activity of telomerase were examined by the method of TRAP, Results: ( 1 ) After treatment with malate at various concentration for 48 h, malate decreased telomerase activity, The maximum down-regulation was obtained at a concentration of 10μmol/L, A time course experiment of telomerase activity in Hela cells treated with 10 μmol/L malate revealed decreased level as early as 8 h after treatment beginning and down to approximately 11% as contrast to the control level at 48 h. (2) After treatment with GABA at various concentration for 48 h, GABA decreased telomerase activity, The maximum down-regulation was obtained at a concentration of 1 mmol/L, A time course experiment of telomerase activity in Hela cells treated with 1 mmol/L GABA revealed decreased level as early as 8 h after treatment beginning and down to approximately 13% as contrast to the control level at 48 h. Conclusion:The telomerase activity of Hela cells can be down-regulated by malate and GABA,
