LM反义RNA对TGFβ刺激下LM受体及FAK表达的影响

摘要 目的明确大肠癌细胞在转化生长因子(transforminggrowthfactorβ,TGFβ)刺激下层粘连蛋白(lami-nin,LM)表达的变化对其受体及粘着斑激酶(focaladhesionkinase,FAK)表达的影响,初步比较FAK的改变与LM受体的相关性。方法采用125I-LM饱和结合剂量检测方法测定LM受体总数量;免疫印迹技术研究FAK表达的变化。结果TGFβ可以刺激LM受体的表达,而且可以被LM反义RNA抑制。用TGFβ刺激FAK蛋白表达的效果不明显,但在LM反义RNA的作用下,FAK的表达在TGFβ刺激下明显减少。结论TGFβ对LM受体及FAK的表达均有一定的调节作用,其中FAK的调节受LM的影响。 Objective To investigate the effect of the expression of laminin on the expression of LM receptor and focal adhesion kinase stimulated by transforming growth factorβ. Methods The laminin binding receptors was identified by ^125I-laminin saturation binding assays. Western blot technique was used to study the expression of FAK at protein level with ECL reagents. Results The expression of LM receptor stimulated by TGFβ was down regulated by the expression of antisence laminin RNA. No change was found in the expression of FAK when stimulated by TGFβ, but its expression was down regulated with the help of laminin antisense RNA. Conclusion The expression of LM receptors and FAK could be regulated when stimulated by TGFβ and LM takes part in the regulation of FAK.
