微卫星多态性检测时银染色和同位素技术比较 被引量:6

COMPARISON OF SILVER STAINING TO ISOTOPIC AUTORADIOGRAPHIC TECHNIQUES IN DETECTING MICROSATELLITE POLYMORPHISMS

摘要 为了比较银染色技术与传统的同位素技术在微卫星多态性检测中的优缺点,对ll8例正常男性基因组DNA,用Y染色体特异性微卫星作聚合酶链式反应,经聚丙烯醚胺序列分析后,分别作银染色和同位素检测。结果表明前者重复性好,结果满意,具有经济、对操作者和环境无害、一天即可出结果等优点。对银染色的不足之处进行了探讨。 Hu Lixian(Department of Nuclear Medicine, Xiangya Hospital, Hunan Medical University)In order to compare the advantages and disadvantages of silver stahhg with traditional radioisotopic .method during microsatellite polymorphism detections,l 18 male normal human ge-nomic DNA were extracted followed by polymerase chain reaction(PCR).After running on poly-acrylamide/bisacrylamide sequencing gel,both silver staining and isotopic detection were per-formed at amplified microsatellite regions derived from human Y chromosome specific DNA(YCAⅡ,YCA Ⅲ and Y-27H39).Giving reproducible and satisfactory results,silver staining de-tection has been shown to be cheaper,less hazardous for operators and environment,ahd lesstime consuniing. Experimental results can beobtained within one day.Some disadvantages on sil-ver staining were also discussed.
