摘要
目的:研究天然红心鸭蛋紫杉紫素对人星形胶质瘤U251细胞增殖、凋亡的干预作用。方法:研究对象为U251人星形胶质瘤细胞系,细胞增殖采用MTT比色法检测,细胞凋亡采用荧光显微镜、激光扫描共聚焦显微镜(LSCM)技术法检测,U251细胞浆中钙离子浓度采用LSCM测定。结果:天然红心鸭蛋紫杉紫素对胶质瘤细胞的生长具有明显的抑制作用,使用小剂量(0.25μmol·L-1)的紫杉紫素能有效地诱导胶质瘤细胞凋亡,并显著提高胶质瘤细胞内Ca2+浓度。结论:天然红心鸭蛋紫杉紫素可以显著抑制U251细胞增殖,且呈时间-效应和剂量-效应依赖关系,同时使U251细胞内Ca2+浓度升高,诱导U251细胞凋亡。
Objective: To study the effects of rhodoxanthin extract from natural red yolk of duck's eggs on growth inhibition and apoptosis of human glioma cells line U251 in vitro. Methods: To observe the effects of rhodoxanthin on proliferation and apoptosis of U251 cells, the MTT, optical microscope, fluorescence and laser scanning confocal microscope (LSCM) were adopted to reveal the cell's morphology. Results: The proliferation of U251 cells was markedly restrained, and the apoptosis of U251 cells were induced by rhodoxanthin with the low, middle and high doses(0.25, 0.5 and 1.0μmol· L^-1 , respectively). At the same time rhodoxanthin inhibited the growth of U251 cells significantly in a dose and time-dependent manner in vitro. The apoptosis of U251 cells were induced effectively by low concentration of rhodoxanthin (0.25μmol·L^-1 ) and in a short time (48 hours). The concentration of Ca^2+ in test group cells remarkably increased. Conclusion: The apoptosis of U251 was induced and the proliferation of U251 ceils were restrained by the rhodoxanthin extract from natural red yolk of duck's eggs. The increase of Ca^2+ concentration in cytoplasm of test group cells was the important cause of Human Glioma U251 apoptosis.
