摘要 目的:观察骨髓间充质干细胞(BMMSC)对急性淋巴细胞白血病(ALL)小鼠异基因骨髓移植(allo-BMT)后移植物抗宿主病(GVHD)和移植物抗白血病(GVL)的影响。方法:以ALL小鼠为动物模型,对其进行al-lo-BMT的同时,静脉输注经体外培养的供鼠BMMSC,同时设立单纯allo-BMT对照组。流式细胞仪检测受鼠CD4+、CD8+T细胞亚群的差异;记录受鼠存活时间;观察受鼠发生GVHD一般反应及病理学变化。结果:BMMSC减少受鼠CD4+T细胞数量的同时增加CD8+T细胞的数量;明显延长ALL小鼠allo-BMT后的生存时间;推迟GVHD发生的时间。结论:BMMSC与allo-BMT联合移植具有一定的GVL;BMMSC能抑制ALL小鼠allo-BMT后GVHD的发生,但同时具有一定程度的GVL。 Objective: To observe the effect of bone marrow mesenchymal stem cells (BMMSCs) on graft versus host disease (GVHD) and graft versus leukemia (GVL) after allogeneic bone marrow transplantation (allo-BMT) in acute lymphocytic leukemia (ALL) mice. Methods: Both bone marrow cells and BMMSCs obtained after three to four weeks of culture from donor mice were transplanted into the recipient mice, which were injected with leukemia cells five days before, and the control group was injected with bone marrow cells alone. The impact of BMMSCs on the quatity of CD^4+ and CD^8+ T cell in vivo after allo-BMT was evaluated by flow cytometry, the survial time after allo-BMT was recorded, and the general manifestation and pathological changes of GVHD was observed. Results: BMMSCs could decrease the quantity of CD^4+ T cells and increase CD^8+ T cells in vivo, BMMSCs could increase the survival time obviously, and delay the onset of GVHD of ALL mice treated with allo-BMT. Conclusion: GVL seems to preserve in the recipient mice injected with bone marrow cells along with donor BMMSCs. BMMSCs could alleviate GVHD and maintain GVI. after allo-BMT in ALL mice.
