摘要
目的:建立以高效液相色谱-荧光法测定布美他尼片中主药含量的方法。方法:色谱柱为KromasilC18,流动相为乙腈-水(50∶50),荧光检测激发波长为228nm、发射波长为418nm,内标为萘啶酸,流速为0.6ml/min,进样量为20μl,柱温为室温。结果:布美他尼检测浓度在16.75~335.00μg/ml范围内线性关系良好(r=0.9997),平均回收率为99.26%(RSD=0.49%,n=3)。结论:本方法可用于布美他尼片中主药的含量测定,且操作简便、结果准确。
OBJECTIVE: To establish a method for content determination of the principal agent in bumetanide tablets by HPLC- fluoremetry.METHODS: HPLC fluoremetry was carried out using a Kromasil C18 column and a mobile phase consisting of acetonitrile- water(50 : 50), the fluorescence detector of 228nm for excitation and 418nm for emission was used,the internal standard was dixiben,the flow rate was 0.6ml/min, the sample size was 20bd, the column temperature was the same as room temperature. RESULTS:The concentration of bumetanide was linear in the range of 16.75-335.00μg/ml( r = 0.9 997), the average recovery was 99.26% (RSD = 0.49%, n = 3) .CONCLUSION:The present method is convenient and reliable,which can be used for content determination of the principal agent in bumetanide tablets.
