摘要
研究发现,苦味酸在还原糖-碳酸钠焦糖化反应中有显著提高其测定灵敏度的作用,确定了最佳反应条件,建立了快速测定植株中微量糖的分光光度法.还原糖含量在0.1~0.5 mg·L-1范围内符合比耳定律,表观摩尔吸光系数ε460=0.37×104L·mol-1·cm-1,回收率在97.0%~109.0%范围内,RSD在1.11%~3.51%之间,蔗糖及果糖对测定无干扰.
It was found that the sensitivity of the carnmel reaction for the determination of reducing sugar was remarkably increased by the addition of picric acid to the reaction mixture. Beer's law was kept in the concentration range of 0. 1-0. 5 mg · L^-1 of reducing sugar. The molar absorptivity was found to be 0. 37× 104L· mol^-1 · cm^-1 at the absorption maxima 460 nm. Values of RSD's (n=5) obtained in the precision tests were in the range of 1.11% to 3.51%, and results of recovery test were in the range of 97. 0% to 109. 0%. Sucrose and fructose were found to have no interference in the determination.
