摘要 目的:研究促性腺激素释放激素激动剂(GnRHa)对体外培养的卵巢黄素化颗粒细胞分泌功能的影响。方法:对16例接受体外受精患者卵泡液中分离的卵巢黄素化颗粒细胞进行体外培养,利用免疫组化法检测颗粒细胞的卵泡刺激素受体(FSHR)蛋白表达鉴定颗粒细胞的纯度,检测加入GnRHa(实验组)体外培养颗粒细胞分泌雌二醇(E2)、孕酮(P)、睾酮(T)、表皮生长因子(EGF)和胰岛素样生长因子1(IGF-1)的水平并与未加GnRHa(对照组)比较。结果:从卵泡液中分离培养的卵巢黄素化颗粒细胞,75%以上呈FSHR蛋白阳性表达;100ng/L的GnRHa对体外培养颗粒细胞作用24h后上清液中每1000个细胞中E2含量为(0.83±0.46)ng/L,明显低于对照组的(2.60±1.53)ng/L(P<0.05);P,T,IGF-1和EGF的含量与对照组差别无统计学意义。结论:GnRHa减弱了颗粒细胞分泌E2的功能,推测GnRHa对卵泡发育和卵母细胞可能有负面影响。 Objective: To study the effects of Gonadotrophin-releasing hormone agonist (GnRHa) on the secrete function of the luteinized granulose cell in vitro. Methods: Human granulosa cells isolated from follicular fluid of 16 patients received IVF or ICSI were cultured in vitro. FSHR expression of granulosa cells were examined by immunobiochemistry assay. The levels of E2, P, T, EGF, and IGF-1 in granulosa cells in GnRHa group were examined and compared with control group. Results: Over 75% granulosa cells isolated from follicular fluid expressed FSHR, After co-cultured with 100 ng/L. GnRHa for 24 h, the value of E2 dropped to (0.83± 0,46) ng/L, which was significantly lower than the control (2.60 ± 1.53) ng/L (P 〈 0.05). There were no difference in P, T, IGF-1, and EGF between these two groups. Conclusion: The secretion of E2 from granulosa cells is diminished by GnRHa, GnRHa maybe has a negative effect on follicles and oocytes.
