氯菊酯农药抗体制备及酶标抗原合成

摘要 为建立一种快速检测氯菊酯的酶免疫学方法,以自制的氯菊酯半抗原(Py)与牛血清白蛋白(BSA)偶联得到免疫原.免疫健康新西兰白兔获得特异性多克隆抗体.用间接酶联免疫吸附法(ELISA)鉴定抗体效价为1:2500和1:4000.合成了辣根过氧化物酶(HRP)标记抗原(HRP-Py),并经紫外分光光度法鉴定,摩尔分子比为2.5:1. To establish a kind of competitive ELISA technique for rapid detecting of permethdn, residue Bovine serum albumin (BSA) was used to couple with self-made semi-antigen permethrin (Py) to obtain immunity source. The complete-antigen BSA-Py acted as immunoantigen to obtain the specific polyclonal antibody. The effect values of antibody were identified through indirect competitive ELISA to be 1:2500 and 1:4000. The HRP labeled antigen (HRP-Py) was synthesized and identified with UV and the molar ratio was 2.5:1.
