摘要
目的:通过对ras相关GTP结合蛋白A(Ras-relatedGTPbindingA,RRAGA)基因的单核苷酸多态(Singlenucleotidepolymorphism,SNP)进行检测,研究RRAGA基因与肺癌的相关性。方法:对RRAGA基因的启动子、外显子区域,使用PCR扩增和DNA测序方法,分别在24例肺癌患者的肺癌组织及24例正常人的外周血进行测序,比较它们之间可能存在的差异,确定该基因是否与肺癌相关。结果:RRAGA基因有4个SNP,其中3个SNP出现在RRAGA基因的启动子区,1个SNP出现在RRAGA基因的3'非翻译区,这4个SNP在正常人与肺癌患者无差异;但这4个SNP与国际公共数据库公布的SNP完全不同。结论:RRAGA基因与肺癌无相关性,中国人RRAGA基因的SNP与外国人的有很大差别。
Objective: To study the association of Ras-related GTP binding A(RRAGA) gene with lung cancer by single nucleotide polymorphism (SNP) detection. Methods: Sequences of coding regions (including sequence near splicing site) of RRAGA gene was detected by PCR-sequencing in 24 lung cancer patients and 24 normal controls. Results: A total of 4 SNPs were identified for RRAGA genel Three SNP located in promoter and one in 3'UTR of RRAGA gene. There was no difference for RRAGA gene between lung cancer and normal control. But the 4 SNP founded were complete different from the SNP publicized in the international database. Conclusion. The RRAGA gene was irrelevant for lung cancer, and the SNP of RRAGA gene was distinct between Chinese and foreigners.
