Toll样受体4在冠心病患者外周血细胞的表达检测及临床意义 被引量:5

Detection of the expression of TLR4 in human leucocytes of the atherosclerotic by flow cytometry and its clinical significance

摘要 目的探讨Toll样受体4(Tolllikereceptor4,TLR4)在外周血白细胞上的表达及其表达差异在动脉粥样硬化中的意义。方法采用流式细胞术检测50例冠状动脉粥样硬化性心脏病患者及40名健康对照者外周血淋巴细胞、单核细胞和中性粒细胞上TLR4的表达;全自动生化分析仪测定所有入选者血脂水平。结果TLR4在外周血淋巴细胞和中性粒细胞的阳性率平均值低于5%;单核细胞的TLR4阳性率在冠心病组为(55.11±16.30)%,显著高于健康对照组的阳性率(42.91±15.70)%(P<0.01);而冠心病血脂正常组和血脂异常组的TLR4阳性率无统计学意义[(54.16±14.60)%;(56.41±18.30)%,P>0.05]。结论TLR4主要表达于外周血单核细胞,在动脉粥样硬化患者中其阳性率升高;而且TLR4的阳性率高低与患者血脂正常与否无关;TLR4及其介导的天然免疫应答与动脉粥样硬化的发生发展存在一定的相关性。 Objective To explore the differential expression of Toll-like receptor 4 (TLR4) in human leukocytes and its significance in atherosclerosis. Methods The positive rates of TLR4 in human lymphocytes, monocytes and polymorphonuclear leukocytes were measured by flow cytometry in 50 patients with coronary arteriosclerosis disease (CAD) and 40 controls without CAD. And the level of plasma lipids were determined by automatic biochemistry analyzer. Results The positive rates of TLR4 in lymphocytes and polymorphonuclear leukocytes were below five percent in both groups, and the positive rate of TLR4 in monocytes in CAD group was significantly higher than that in controls[ (55. 11 ± 16. 30)% versus (42.91 ± 15.70)% ,P〈0.01]. But no significant difference was found in the positive rate of TLR4 between CAD group with normal level of plasma lipids and the CAD group with abnormal level of plasma lipids [ (54. 16 ± 14.60)%versus (56.41 ±18.30)%,P 〉0.05]. Conclusions TLR4 is mainly expressed in human monocytes, and the positive rate of TLR4 is increased in atherosclerotic. And the positive rate of TLR4 has no correlation with the level of plasma lipids. TLR4 and the innate immune response mediated by it are associated with arteriosclerosis.
