模式蛋白分离用的A3-2型空间连续自由流电泳仪

摘要 目的研究空间微重力环境对电泳分离的影响,证实自主研制的A3-2型电泳仪的设计和工艺,为我国空间电泳研究的发展和生物材料加工及空间制药奠定基础。方法用A3-2型连续自由流电泳仪,分别在地面和空间对细胞色素C和牛血红蛋白两种模式蛋白进行电泳分离实验,对其结果进行分析比较。结果电泳仪在“神舟”4号飞船上成功地进行了空间电泳分离实验,并获得了完整的飞行试验数据。返回舱着陆后,回收的电泳仪装置完好无损,得到了全部(30管)分离产物。结论在线光电检测及分离产物的检测结果都说明,空间微重力环境下的电泳分离效果明显优于地面的结果。飞行试验表明,A3-2电泳仪的结构设计和工艺是正确和可行的,它为我国首次进行的空间电泳分离实验取得圆满成功提供了保障。 Objective To study the influence of space microgravity environment on the electrophoresis separation process and to qualify the design and technologies of A3-2 device. Method A3-2 device was flown on board the recoverable capsule of Shenzhou -Ⅳ spacecraft and the electrophoretic separating experiments of the model proteins, Bovine Hemoglobin and Cytochrome C were performed. Result All of the engineering and scientific parameters in the experiment were obtained. Everything in the recovered electrophoresis unit was fine and 30 fractions were got completely after landing. Both of the results obtained by on-line LED detector and SP22 UV spectrophotometer from the separated products, showed that the separation quality of proteins by the continuous free-flow electrophoresis device was better under microgravity than on the ground. Conclusion The A3-2 device can work well in space. It ensures the success of space electrophoresis separating experiment for the first time in China.
