高效液相色谱法测定华细辛中马兜铃酸A含量

Quantitative analysis of aristolochic acid A in Asarum sieboldii by HPLC

目的测定华细辛中马兜铃酸A的含量。方法建立高效液相色谱方法,采用Agilent Zorbax SB-C_(18)色谱柱(4.6mm×250mm,5μm),以乙腈-1%冰醋酸水溶液(39:61)为流动相,流速为0.8mL·min^(-1),检测波长为317nm,分别测定20个华细辛样品中马兜铃酸A的含量。结果高效液相色谱法的回收率为98.9%,RSD为2.1%,标准曲线回归方程为y=2.40x+0.267, 在5.2～68ng内呈良好线性关系。结论华细辛中马兜铃酸A含量在6.8～45.4μg·g^(-1)。

OBJECTIVE To determine aristolochic acid A in Asarum sieboldii Miq. METHODS A HPLC method was established with a Agilent Zorbax SB - C18 column （4.6 mm× 250 mm, 5 μm）, a mobile phase of acetonitrile-0.1% acetic acid solution （39：61）, a flow rate of 0.8 mL· min^- 1 and a detection wavelength of 317 nm in order to determine the contents of aristolochic acid A in the 20 samples of Asarum sieboldii Miq.. RESULTS The average recovery rate was 98.9 %, RSD was 2.1%, and the calibration curve was y = 2.40x ＋ 0. 267 with a linear range of 5.2 ng to 68 ng. CONCLUSION The concentration of aristolochic acid A is between 6.8 and 45.4μg· g^-1.
