绿豆芽超氧物歧化酶的纯化及性质研究 被引量:2

摘要 采用硫酸铵盐析、超滤、DEAE-SephadexA-50柱层析,从绿豆芽中提纯了超氧物歧化酶。酶的纯化程度达到每mg蛋白含3194.7U,总酶活回收率为38.9,纯化倍数为30,7.酶理化性质研究表明,所得酶在270nm紫外光区有一吸收峰;聚丙烯酰胺凝胶电泳显示2条谱带;氨基酸分析表明含308个氨基酸残基;在60℃处理40min,酶活损失很小;在pH2~11范围活性稳定;KCN、EDTA对酶活性具抑制作用。 Sod from green bean sprouts was purified through(NH4)2SO4.solt-surplussion,ultrafiltration and DEAE-sephadex A 50.The puration exhibited 3 1 94.7(L/protein activity),thetotal recovery rate of enzyme activity was 38.9 and the purification times was 30.7.The studyon its physicochemical properties showed that the ready-owned enzyme had a absorption peakat 270 nm in ultraviolet spectrum and two derivative spectra were observed through polyacry-lamide get electrophoresis.The analysis of.amino acid showed that it had 308 amino acidresidues and the enzyme activity loss was little when treated at 60 celsius for 40 minutes.Between pH2 and pH11,it had a stable activity but could be inhibited by KCN,EDTA.
