摘要
目的研究氡及其子体吸入对大鼠肺与血细胞的氧化应激损伤。方法雄性Wistar大鼠20只,随机分为4组,除对照组外,处理组整体暴露于多功能生态氡室,吸入氡及其子体的累积剂量分别达27,52,105工作水平月(Working Level Month,WLM)后,检测各组大鼠肺组织匀浆以及外周血的超氧化物歧化酶(SOD)、丙二醛(MDA)含量,并使用荧光探针技术,检测外周血单个核细胞(Peripheral Blood Mononuclear Cell,PBMC)以及支气管肺泡灌洗液(Bronchoalveolar Lavage Fluid,BALF)细胞,其细胞内活性氧(Reactive Oxygen Species,ROS)的平均含量。结果与对照组相比,各处理组SOD含量均有降低(P<0.01),MDA均有上升(P<0.01),其中肺组织匀浆中SOD含量,27WLM组与105WLM组有显著性差异(P<0.01),MDA含量27WLM组、52WLM组与105WLM组相比分别有显著性差异(P<0.01),外周血中MDA含量,27WLM与105WLM有显著性差异(P<0.01);BALF以及PBMC细胞内的ROS含量,各处理组与对照组相比均增高(P<0.01),但各组间无显著性差异(P<0.05)。结论在该实验条件下,氡及其子体吸入扰乱了大鼠的氧化应激平衡体系,使其受到一定的损伤。
Objective To study the redox damage of lung and blood cells in rats inhaled radon and its progeny. Methods Male Wistar rats were exposed to radon and its peogeny with the cumulative dose up to 27, 52,105 working level month(WLM) respectively. The lever of SOD(superoxide dismutase) and MDA(malondialdehyde) in blood , BALF and lung tissue were investigated. The change of intracellular ROS content in BALF (Bronchoalveolar lavage fluid) cell and PBMC(Peripheral blood mononuclear cell) were also investigated. Result The lever of SOD in blood and lung tissues significantly decreased and the lever of MDA increased. The content of intraeellular ROS in BALF cell and PBMC also increased remarkably. Conclusion The inhallation of radon and its progeny can lead to the persistent disturbance of the redox state in rats.
