上皮钙黏附素/β连环素在滑膜肉瘤中的表达及意义

Expressions and Significance of Ecadherin and β-catenin in Synovial Sarcoma

目的通过研究上皮钙黏附素(E cadherin),β-连环素(-βcaten in)以及k i-67在滑膜肉瘤中的表达变化,探讨上皮钙黏附素/β-连环素(E-cadherin/-βcaten in,E-cad/cat)复合物与滑膜肉瘤的形态发生学及其进展的关系。方法采用免疫组织化学方法(Env is ion二步法)检测E cadherin,-βcaten in以及k i-6 7在2 6例滑膜肉瘤(其中双相分化型6例,单相分化型20例)中的表达情况。结果a)E-cadherin在7例滑膜肉瘤中表达,阳性表达率为26.9%(7/26),且主要表达在有上皮样分化区域的肿瘤细胞膜上,其表达与肿瘤的分型有关;b)-βcaten in除表达于肿瘤细胞细胞膜上以外,57.7%(15/26)出现有意义的异常表达(表现为肿瘤细胞细胞浆或细胞核表达),其异常表达与k i-67指数具有相关性(P<0.01)。结论a)E cadherin/-βcaten in介导了滑膜肉瘤中上皮样分化肿瘤细胞间的粘附,可能参与了该肿瘤的形态学发生;b)-βcaten in的异常表达可能激活了相关信号传导通路,引起滑膜肉瘤肿瘤细胞的异常增殖,从而促进该肿瘤的发生、发展。

Objective To investigate the expression of Ecadherin, βcatenin and ki67 in synovial sarcoma, and to explore the relationship between the expressions and morphogenesis, progress in synovial sarcoma. Methods The Envision immunohistochemical method was used to detect the expressions of Ecadherin, βcatenin and ki67 in 26 cases of synovial sarcoma. Results a）Expression of Ecadherin was localized at the cellular membrane of seven specimens, especially in epithelial components with biphasic synovial sarcomas; b）βcatenin was observed at the cell boundaries and showed wide spread intracytoplasmic or aberrant nuclear expression. There was a correlation between widespread aberrant staining of βcatenin and the ki67 index（P〈0.01）. Conclusion a）The results suggested that Ecadherin/β-catenin plays an important role in cell＇s adhesion, especially in epithelial differentiation, and might t.ake part in the morphogenesis of synovial sarcoma; b）The aberrant cytoplasm/nucleus accumulations of β-catenin might take part in the abnormal proliferation of the tumor cells and the development and progression of synovial sarcoma by activating some kinds of signal pathway.
