Establishment of A Mouse Model for Neisseria gonorrhoeae Infection

摘要 目的尝试建立淋球菌感染动物模型,为研究淋球菌致病机制提供良好的动物模型。方法采用野生株淋球菌FA1090接种雌二醇预处理的雌性BALB-c小鼠,小鼠生殖道拭子接种WGC选择培养基培养分离淋球菌,取小鼠阴道分泌物涂片Gram染色观察有无淋球菌,并用瑞氏染色观察小鼠阴道分泌物中多形核白细胞(polymorphonuclearleukocytes,PMN)的数目。结果受试小鼠生殖道中淋球菌平均存活了8d(2×106CFU接种量),阴道分泌物涂片Gram染色可见淋球菌粘附在上皮细胞上,88%的受试小鼠发生了炎症。结论成功建立了淋球菌感染的动物模型,为研究淋球菌的致病机制、毒力及抗菌药物的研究奠定了基础。 Objective To construct a mouse model for Nelsseria gonorrhoeae genital infection. Methods Female BALB/c mice were treated with 17-β-estradiol and inoculated intravaglnaUy with wild type gonococcus FA1090. The genital swab were verified by the WGC agar plate. The vaginal excretes smears were stained with wright stain and viewed under light microscopy to determine the percentage of PMNs among 100 vaginal cells. Results Survival of gonococci in the animal genital tract continued for an average of 8 days. The stained vaginal smears showed that the pathogens adhered to the epithelial cells and neutrophils.Inflammation occurred in over 88% of infected mice. Conclusion A mouse model for Neisseria gonorrhoeae infection was constructured successfully, which may be used for the study of pathogenicity, virulence and treatment of Neisseria gonorrhoeae infection.
