摘要
目的建立薄层扫描法对华山参中总东莨菪内酯的含量测定方法.方法采用高效硅胶G薄层色谱板,以氯仿-甲醇(15∶1)为展开剂,采用单波长荧光线性扫描法,选择'K400'为滤光片,激发波长340 nm测定.结果东莨菪内酯在0.06~0.3 μg线性关系良好,样品回收率104.40%.结论该方法快速简捷,精密度高,重现性好,可用于华山参中总东莨菪内酯含量的测定.
OBJECTIVE To establish a method to determine the contents of scopoletin in Physochlaine infundibularis Kuang by TLCS. METHODS Single-wavelength TLC scanning was used with chlorofrom-methanol (15 : 1) as developer. The plate was scanned at 340 nm in the fluorescence linear scan mode. RESULTS The linearity was good within the range 0. 06-0. 3μg (r=0. 999 7) . The average recovery was 104. 40%. CONCLUSIONS The method is simple, rapid, sensitive and highly reproducible for detecting scopoletin in Physochlaine infundibularis Kuang.
