维生素K_2对NB4细胞生长、凋亡、周期阻滞及分化的诱导作用

摘要 目的研究维生素K2(VK2)对NB4细胞生长、凋亡、周期阻滞及分化的诱导作用。方法通过四唑盐比色法、形态学观察、流式细胞仪检测细胞早期凋亡率、细胞周期分析、NBT还原法和流式检测CD14、CD11b阳性率研究维生素K2对NB4细胞的影响。结果NB4细胞经VK2作用后增殖受到抑制;VK2作用NB4细胞72h后,细胞形态呈现凋亡特征,细胞早期凋亡率随浓度升高而升高,与对照组比较及两两比较P<0.01;各药加组G0/G1期细胞均较空白对照组升高,各加药组与空白对照组比较P<0.01,但各加药组之间两两比较无统计学意义;各加药组NBT阳性率、CD14、CD11b表达均较空白对照组无统计学意义。结论VK2对NB4细胞有生长抑制作用,且具有时间及剂量依赖性;VK2可诱导NB4细胞凋亡,且呈剂量依赖性;VK2可诱导NB4细胞周期受阻于G0/G1期,但细胞周期阻滞作用无量效关系;VK2无单独诱导NB4细胞分化作用。 Objective To study the effect of vitamin K2(VK2)on the growth, apoptosis, arrest of cell cycle and differentiation of human leukemia acute promylocytic leukemia(APL) cell line NB4. Methods We examined the effects of growth of cells by MTT test, and observed the changes of morphologic features of cells, and examined the effects of differentiation by NBT test. The rate of apoptosis, cell cycle and expression of CDl4 and CDnb was detected by flow cy- tometry. Results Vitamin K2 can inhibit the proliferation of NB4 cells. The early apoptosis percentage of NB4 ceils incubated with VK2 for 72h was higher than those in control groups ( P 〈 0.01) and this effect shows dosage-dependent relation. Cells in G0/G1-phase of NB4 ceils incubated with VK2 for 72h were higher than those in control groups ( P 〈 0.01), but this effect didn't show dosage-dependent relation. After incubated with VK2 for 72h, NBT positive ceils percentage and expression of CDI4 and CDIIb show on statistical differences with those in control groups (P 〉 0.05). Condusions VK2 can inhibit NB4 ceils proliferation and this effect shows dosage and time-dependent relation. VK2 can induce NB4 cells apoptosis and this effect shows dosage-dependent relation. VK2 can interfere the cell-cycle phase in NB4 ceils and cause cell remain in G0/G1-phase but this effect doesn't show dosage-dependent, relation. VK2 can't induce NB4 ceils differentiation alone.
