摘要
目的观察炎性痛条件下西非单叶豆同工凝集素(BSI-B4,可以选择性标记参与痛觉信息传递的C纤维及其终末)结合位点的变化情况以及电针对其调节。方法采用大鼠单侧佐剂性关节炎性痛模型,采用免疫荧光组织化学技术观察了致炎后不同时间段(分别取第3、7d)背根节BSI-B4结合位点的表达情况,以及在针刺(电针环跳穴、阳陵泉穴)条件下该指标的变化情况。结果注射后第3、7d各组大鼠背根节的BSI-B4结合位点表达关系比较如下:炎性痛组(包括3、7d组)>电针组(包括3、7d组)>生理盐水组(包括3、7d组)(P<0.01)。且炎性痛组大鼠BSI-B4表达还表现为3d组>7d组(P<0.05)。结论电针可以下调炎性痛大鼠背根节BSI-B4结合位点表达的增多,这可能是电针镇痛的机理之一,即通过减缓炎性痛大鼠C伤害性感受器的持久发放达到治疗炎性痛的目的。
Objective To observe the changes of Banderaea simplicifolia Isolectin IM (BSI-IM) binding sites expression in the dorsal root ganglion (DRG) following inflammatory pain and elucidate the influence of electroacupuncture (EA) to it. Methods In this study, we applied the complete Freund' s adjuvant (CFA)-induced inflammatory pain model. In addition, BSI-B4 binding sites were detected in DRG by using immunofluresence histochemistry following CFA-induced inflammation and EA treatment. Results At both 3^rd day and 7^th day following injection, the mean immunoreactive area and optical density of BSI-B4 labeled products, respectively, all showed that: inflammatory group 〉 EA treatment group 〉 control group (all P 〈 0.01). And in inflammatory group, the expression of N BSFIM also showed that: 3^rd day group 〉 7^th day group (P 〈 0.05). Conclusion The above data indicated that EA could down-regulate the BSI-B4 binding sites expression in DRG . It maybe one of the mechanism of EA analgesia that was probably to weaken the discharge of unmyelinated nociceptive afferent terminals which contributed significantly to the hyperalgesia.
