摘要
目的:探讨老龄小鼠肝线粒体结构和功能的增龄性改变和红花黄色素(Safflower Yellow Pigment,SYP)对肝线粒体抗氧化损伤的保护作用。方法:以衰老小鼠为研究对象,测定了肝线粒体丙二醛(MDA)含量,Mn—超氧化物歧化酶(Mn—SOD)活性,Ca^(2+)—ATPase 活性、复合体Ⅰ及复合体Ⅱ+Ⅲ活性,观察不同剂量 SYP 对上述各指标的影响。结果:老年组肝线粒体 Mn—SOD 活性、Ca^(2+)—ATPase 活性、复活体Ⅰ和复合体Ⅱ+Ⅲ活性显著降低,而 MDA 含量升高;SYP 大、中剂量组可升高 Mn—SOD、Ca^(2+)—ATPase、复合体Ⅰ及复合体Ⅱ+Ⅲ活性。结论:红花黄色素通过提高机体抗氧化能力、保护复合体酶活性、改善细胞内钙离子超载而起到延缓衰老的作用。
Objective:To investigate the age-related changes of hepatocytic mitochondria injury and the protective effects of Safflower Yellow Pigment(SYP)on the mitochondria in senile model mice.Methods:The activities of Mn- SOD,Ca^(2+)-ATPase,complex Ⅰ,complex Ⅱ+Ⅲ in mitochondria of hepatocytic and the MDA content were measured. The effects of different doses SYP on the above-mentioned indices were studied.Results:The activities of Mn-SOD, Ca^(2+)-ATPase,complex Ⅰ,complex Ⅱ add Ⅲ in senile model group were lower,but the level of MDA was significant higher than that of young and middle group.SYP groups had evidently heightened the activities of Mn-SOD,Ca^(2+)- ATPase,complex Ⅰ,complex Ⅱ+Ⅲ,the lower contents of MDA were found in SYP groups.Conclusion:SYP has the antioxidative effect by diminishing mitocbondria lipid peroxidation and improving the activities of complex and overcharging of cell Ca^(2+).
