高三尖杉酯碱对Jurkat细胞端粒酶活性的影响及机制研究 被引量:2

摘要 目的探讨高三尖杉酯碱(HHT)对人T淋巴细胞白血病细胞株Jurkat细胞端粒酶活性的影响及其机制。方法采用改良的K rupp荧光素标记的端粒重复扩增(TRAP)方法检测HHT对Jurkat细胞端粒酶活性的影响。半定量RT-PCR方法检测HHT作用后Jurkat细胞端粒酶逆转录酶(hTERT)及c-m yc原癌基因mRNA的表达水平。结果HHT对Jurkat细胞端粒酶活性及hTERTmRNA、c-m yc基因mRNA表达有明显抑制作用。结论HHT可通过下调hTERT基因转录抑制淋巴细胞的端粒酶活性。c-m yc可能参与调控淋巴细胞hTERT基因转录。 Objective To investigate the effect and mechanism of homoharringtonine on telomerase activity of human T lymphocyte Jurkat cells. Methods The telomerase activity of Jurkat cells was examined by modified Krupp TRAP assay. The expression of c-myc and hTERT mRNA was half-quantified using RT-PCR assay. Results Telomerase activity was significantly inhibited by HHT. Simultaneously,the down-regulation of hTERT mRNA and c-myc mRNA expressions was observed in Jurkat cells treated with HHT. Conclusion The inhibition of telomerase activity by HHT is due to the repressed hTERT mRNA expression, c-myc gene might regulate the transactivation of hTERT gene in the cells.
