The Effect of Triptolide on MCP-1 mRNA Expression in Wistar Rats with Experimental Autoimmune Encephalomyelitis

摘要 目的探讨雷公藤内脂醇(TRD)对EAE大鼠MCP-1mRNA表达的影响。方法将Wistar大鼠免疫后7d随机分成4组:EAE组、TRD1组、TRD2组、TRD3组,给予不同剂量的药物注射,取脑和脊髓制成石蜡切片,进行HE和MCP-1mRNA的原位杂交染色,比较各项临床指标。结果TRD在每只鼠每日40g剂量下影响EAE大鼠MCP-1mRNA的表达。结论雷公藤内脂醇可增加循环中的糖皮质激素从而抑制EAE中MCP-1mRNA的表达,抑制EAE。 Objective To study the immunosuppressive effects of triptolide to MCP-1 mRNA expression of EAE Wistar rots. Methods GPSCH, CFA and PT-immtmized Wistar rats are divided randomly into four groups, group EAE, group TRD 1, group TRD 2 and group TRD 3. The brains and spinal cords were removed for HE staining and in situ hybridizations (ISH) staining. Results Group TRD 3 (40μg TRD · rat^-1 · d^-1) implies a therapeutic effect. Conclusion TRD arrests the development of EAE in rats, partly through increasing circulating glucocorticoid and down-regulating the expression of MCP-1 mRNA.
