摘要
目的检测C3A细胞在正常人和在慢性重型肝炎血浆中解毒功能及膜的完整性,探讨慢性重型肝炎患者血浆在体外对C3A细胞株功能的影响。方法把C3A细胞分为两组,一组用慢性重型肝炎患者血浆(100%CSHP)培养,另一组用正常人血浆(100%NHP)培养。分别于第24h及第48h,检测培养细胞上清液中丙氨酸氨基转移酶(ALT)及天冬氨酸氨基转移酶(AST)含量,于第48h检测C3A细胞内还原型谷胱甘肽(GSH)含量。结果在100%CSHP中和100%NHP培养第24h及第48h,培养细胞上清液中ALT和AST含量,100%CSHP组与对照组间差异有非常显著性意义(P<0·01),且第48h与第24h含量间差异有显著性意义(P<0·05)。分别检测在100%CSHP中和100%NHP培养第48h的C3A细胞内GSH含量,100%CSHP组与对照组间差异有显著性意义(P<0·01)。结论慢性重型肝炎患者血浆使C3A细胞内GSH消耗明显增加,导致其解毒功能下降;CSHP使C3A细胞膜受损,并随时间延长而逐渐加重。
Objective Through comparing the detoxic function and the complete membrane of C3A cells in plasma of normal people and chronic severe hepatitis, to investigate the effects of plasma from patients with chronic severe hepatitis on the detoxic function and membrane's integrity of the C3A cells in vitro. Methods C3A cells were divided into two groups which were separately cultured in NHP and CSHP. Detecting the content of ALT and AST was detected in the superant after being cultured at the time of 24 hour and 48 hour. The content of glutathione (GSH) was detected inside the cells after being cultured for 48 hour. Results The contents of ALT and AST at the period of the 24 h or the 48 h were much higher than those in NHP. The difference was statistically significant ( P 〈0. 01 ). The content is different at 48 h and 24 h ( P 〈0. 05). The content of GSH at the period of 48 h was lower than that in NHP. The difference was statistically significant ( P 〈0. 01 ). Conclusion CSHP can not only decrease the detoxic capability of C3A cells by exhausting GSH inside the cells, but also damage the membrane integrity by time and aggravation.
