The gene therapy of cancer is a cancer-treatment technique by expressing anticancer genes in tumor cells by rationally designed vectors. Nowadays, more and more crucial molecules in tumor cells have been identified that are related to the unlimited proliferation of cancer cells. Some can inhibit the apoptosis, some can promote angiogenesis, and some can help tumor cell evade immune surveillance. These molecules are ideal targets in future cancer gene therapy. This article reviews the features and the application of two important molecules in tumor cells: hypoxia-inducible factor 1 ( HIF-1 ), signal transducer and activator of transcription 3/5 (STAT3/5)
