反相液相色谱法测定小檗碱型生物碱

RP-HPLC determination of the berberine type alkaloids from the Rhizoma coptidis

建立反相液相色谱法,测定20%醋酸提取的黄连药材中小檗碱型生物碱的方法.色谱柱为Permaphase ODS柱,流动相为甲醇-硼酸盐缓冲液(0.02 mol/L硼砂,用硼酸调节pH=8.5)(80∶20),内含0.2 mL二乙醇胺,波长为254 nm,外标法测定峰高.在以上条件下,小檗碱、小檗胺、巴马汀和药根碱可以较好分离.线性回归方程为Y=25 214.14X,R=0.999 3,平均回收率为99.485%.测定方法简单,快速,结果较满意.也适用于其它小檗碱型生物碱的分离和测定.

A reversed-phase HPLC method was developed for the quantitative determination of Berberine type alkaloids form Rhizoma coptidis by 20 % acetic acid. An ODS column was used, with the mobile phase consisted of methyl alcohol and borax-boric acid mixture （ 0.02 mol/L borax, adjust pH to 8.5 with boric acid ） （80：20） ,containing 0.2 mL two ethanol amine, the detecting wave-length is a 254 nm . Four kinds of berberine type alkaloids, berberine, berbamine, palmatine, jatrorrhizine can be separated completely. The regression equation is Y=25214.14X R= 0. 9993 ,and the average recovery is 99.485 %. The method can be used for the research of Rhizoma coptidis and for the routine analysis of the crude drugs and Chinese traditional medicine containing berberine.
