应用单管PCR扩增测序分型法分析成都汉族HLA-DRB1基因多态性

摘要 目的应用DNA分型技术分析成都汉族个体HLA-DRB1等位基因分布频率,为进一步研究HLA-DRB1与疾病的关联提供背景资料。方法设计合成引物,建立HLA-DRB1单管PCR扩增测序分型法,对90名无血缘关系的健康个体进行HLA-DRB1基因分型。结果共检出39种HLA-DRB1等位基因,基因频率较高的有HLA-DRB10901(26.11%),1202(7.22%),同时检测出HLA-DRB1AHAW、HLA-DRB1DRw12、HLA-DRB1w13b等少见等位基因。统计分析表明该群体HLA-DRB1位点的基因型分布符合HardyWeinburg平衡(χ2=806.46,df=780,P>0.05)。结论证实了单管PCR扩增测序分型法用于HLA-DRB1基因分型的可靠性和适用性,并提供了成都汉族群体的基因分布频率资料。 Objective To analyze the HLA-DRB1 allele distributing frequency of individual of Hans in Chengdu with sequence based typing and to provide background data for further research about the relationship between HLA-DRB1 and diseases.Methods The technique of Single-tube-PCR amplification assays was adopted to type the HLA-DRB1 genotypes of 90 healthy individuals of Hans in Chengdu. Results Thirty-nine HLA-DRB1 alleles were observed.The frequency of HLA-DRB1* 0901 (26. 11% ), 1202(7.22% ) was higher than that of the others,and such infrequent alleles as HLA-DRB1 * AHAW,HLA-DRB1* DRw12,HLA-DRB1 * w13b were also found. Statistical analysis showed that HLA-DRB 1 allele distributing accorded with the Hardy Weinburg balance (x^2 = 806.46, df = 780, P〉0.05 ).Conclusion It is proved that the Single-tube-PCR amplification assays is reliable and available to determine HLA-DRB1 genotype, and the HLA-DRB1 allele distributing frequency data of individuals of Hans in Chengdu were provided.
