摘要 基于DNA与RNA对中药葛根素的荧光有猝灭作用而建立了DNA、RNA的测定新方法.在生理pH值附近,当激发波长为266 nm,葛根素在460 nm处出现荧光,随着核酸的加入,葛根素的荧光强度线性下降,本文对4种不同核酸的线性方程、线性范围、检测限进行了测定,对外来离子的干扰进行了研究,与经典的核酸测定方法进行了对比,并对核酸使葛根素荧光猝灭作用机理进行了探讨. A novel fluorescence method has been developed for rapid determination of DNA or RNA with puerarin as a fluorescence probe, based on the fluorescence quenching of puerarin in the presence of DNA or RNA. The maximum excitation and emission wavelengths are at 266 nm and 460 nm respectively at physiological pH. Under optimal conditions, the calibration graphs, linear ranges and the corresponding detection limits were determined. Interferences from foreign substances were tested. Other methods for nucleic acid determinations were compared with this method. The mechanism for the binding of puerarin to DNA was also studied.
