摘要
[目的]观察维生素K3(VK3)对人卵巢癌细胞株SKOV3增殖和凋亡的影响,并探讨其作用机制。[方法]四甲基偶氮唑蓝(MTT)法观察VK3对SKOV3细胞的抑制作用,AnnexinV/PI法测定细胞凋亡,RT-PCR法检测survivin基因的表达。[结果]不同浓度VK3(2、5、10、20μmol/L)作用SKOV3细胞48h后,生长抑制率分别为14.30%,28.65%,44.58%,68.33%。不同浓度VK3(0、2、5、10、20μmol/L)作用48h后,SKOV3细胞的凋亡率分别是4.36%,9.87%,10.80%,29.65%,76.80%。VK3作用后,survivin基因在SKOV3细胞内的表达明显下降。[结论]VK3能抑制SKOV3细胞增殖,并诱导其发生调亡,其作用机制可能与下调survivin基因表达有关。
[Purpose] To observe the effects of Vitamin K3(VK3) on cell growth and apoptosis in human ovarian cancer cell line SKOV3 and to explore its mechanism. [Methods]The inhibitory effect of VK3 on SKOV3 cells was tested by methyl thiazolyl tetrazolium (MTT) method. The rate of apoptotic cell was quantified by Annexin V/PI assay. The change of survivin gene expression was detected by RT-PCR. [Results] The inhibitory rate of VK3 (at concentrations of 0,2,5,10,20μmol/L for 48h) on SKOV3 growth was 14.30%, 28.65%, 44.58%, 68.33% respectively. The apoptotic cell rate was 4.36%, 9.87%, 10.80%, 29.65%, 76.80% respectively after SKOV3 cells were treated with VK3 for 48h at concentrations of 0, 2, 5, 10, 20μmol/L. After treatment with VK3, survivin expression in SKOV3 was down-regulated. [Conclusion ] VK3 can inhibit the proliferation of SKOV3 cells and induce apoptosis, its mechanism may be related to down-regulating survivin gene expression.
