内皮素含量检测在人工血管预衬基质筛选中的意义

摘要 目的研究不同预衬基质对内皮细胞产生内皮素量的影响,探讨内皮素含量检测在预衬基质筛选中的意义。方法应用体外细胞分离、培养、放免GAMMA计数测定等技术,对预衬基质明胶、纤维结合素、血浆和止血灵与血管内皮细胞生长过程中产生内皮素量的关系进行研究,评价内皮素作为反映内皮细胞功能及受损程度指标的可行性。结果不同的预衬基质对内皮细胞产生内皮素的量影响不同,内皮素产生量由低到高为2%明胶、血浆、纤维结合素、1%止血灵。内皮素在人工血管预衬基质的筛选中,可表达预衬基质对内皮细胞功能状态及损伤程度的影响。结论内皮素产生量在细胞培养过程与内皮细胞的损伤程度及功能状态有关,结合细胞计数可完善预衬基质的筛选指标。 Objective To investigate the significance of screening matrix component of blood vessel prosthesis according to endothelin level. Methods The endothelin content was measured in different cultured fluids using 2% gelatin, plasma, fibre-nectin and 1% zhixueling as matrix respectively by radioimmunoassay. The possibility of endothelin content as an index for the damage degree of endothelial cells was assessed. Results Influence of matrix components on endothelin content differed significantly; in the order of the endothelin content formed, the matrix components were 2% gelatin, plasma, fibre-nectin and 1% zhixueling respectively. Endothelin content was associated with function condition and the degree of damage of endothelial cells. Conclusions As an important indication, endothelin content can reflect function condition and damage degree of endothelial cells, and it can be used to screen matrix components of blood vessel prosthesis.
