Establishment of a high-frequency plant regeneration system using Stylosanthes guianensis

摘要 对热研2号柱花草无菌实生苗的胚轴与叶片切段培养进行了研究.结果表明,在MS + 0.5~2.5 mg/L BA均可实现100%的再生频率,其中叶片切段在MS + 1.5 mg/L BA上单位外植体产生的丛生芽数最多,为18.45;而胚轴切段在MS + 2.0 mg/L BA上单位外植体产生的丛生芽数最多,为15.30.经过4周的壮苗培养,切下单芽进行生根培养.最适生根培养基为1/2 MS + 1.0 mg/L IBA + 0.5 mg/L IAA,生根率可达95.84%. The morphogenetie potential of such explants as leaf pieces and hypoeotyl segments from aseptic seedlings of Stylosanthes guianensis cv. Reyan 2 was investigated in this study. The result showed that high-frequency plant regeneration was achieved using MS + 0.5-2.5 mg/L BA with leaf pieces and hypocotyl segments used as explants. The highest shoot number produced per explant of leaf pieces (18.45) and hypocotyl segments (15.30) was obtained on MS + 1.5 mg/L BA and MS + 2.0 mg/L BA,respectively. After 4 weeks of elongation culture on MS +0. 1 mg/L BA,micro-shoots were cut and rooted onto rooting media. The best rooting medium was found to be a half-strength MS supplemented with 1.0 mg/L IBA and 0.5 mg/L IAA.
