摘要
[目的]研究轮叶党参醇提取物水溶部分(WACL)对环磷酰胺(CP)所致突变的拮抗作用。[方法]将大鼠外周血淋巴细胞体外培养,观察在相同的CP浓度(20μg/ml)、不同的WACL浓度(低:0.25mg/ml,中低:0.50mg/ml,中: 1.00mg/ml,中高:2.00mg/ml,高:4.00mg/ml);以及在相同的CP浓度、相同的WACL浓度下,不同的添加WACL和CP 秩序间(方法一:先加CP,后加WACL;方法二:先加WACL,后加CP;方法三:同时加入WACL和CP)的双核徽核率。用胞质分裂阻滞微核测试法(CB—MNT)测定。[结果]①3种处理方法的WACL各浓度都具有抗突变作用,且均呈剂量-效应关系,相关系数r为:-0.744^-0.822,P值均<0.01;②在同-WACL浓度下,比较不同的处理方法间的双核微核率发现:在中、中高、高浓度下,3种处理方法间的双核微核率差异无统计学意义;低浓度和中低浓度时,3种处理方法间双核微核率差异有统计学意义(F=7.75和7.82,P<0.05),先加入WACL者,细胞微核率较低。[结论]WACL具有抗突变和去突变效果;并且在WACL浓度较低时,先加入WACL的抗突变效果较佳。
[ Objective ] To study the antagonistic effect of the water portion of alcohol extract of Codonopsis lanceolata ( WACL ) against CP-induced mutation. [ Methods ] According to cytoplasm-blocked micronuclei test( CB-MNT ) for the peripheral blood lymphocytes in rats, in vitro lymphocytes culture was applied. The rate of micronuclei in binucleated lymphocytes of various concentrations of the water portion of alcohol extract, and rate of micronuclei between various groups defined by different order of adding the water portion of extract and cyclophophamide with different concentrations ( 0.25, 0.50, 1.00, 2.00 or 4.00 mg/ml )of WACL, while CP concentration( 20μg/ml )was fixed. [ Results ] ① Various concentrations of WACL using the 3 kinds of designed methods expressed the antimutagenic effect against CP-induced mutation, and has a dose-response relationship, the correlation coofficient r=-0.744 — -0.822, all of the P 〈 0.01. ② Compared the three groups, when the WACL is at the same level such the middle, middlehigh, high level of the WACL, there is no statistic difference between the 3 methods of design, but at low, low-middle level , there is a statistic difference between the 3 methods of design. When low, the statistic result is F=7.75, P 〈 0.05. When low-middle, the statistic result is F=7.82, P 〈 0.05 ; [ Conclusion ] The WACL has the antimutagenic effect against CP-induced mutation, and it expresses a better effect when adding WACL before CP.
