接骨木中的酚酸类化合物及其对大鼠类成骨细胞UMR106增殖及分化的影响 被引量:25

摘要 目的研究接骨木Sambucus williamsii Hance中抗骨质疏松的活性成分,及其对UMR106类成骨细胞增殖和分化的影响。方法以大鼠类成骨细胞UMR106的增殖为指导活性成分分离的指标,利用各种分离手段追踪分离接骨木茎枝60%乙醇提取物活性部位的化学成分,并利用光谱学方法鉴定其结构,检测对UMR106细胞增殖和分化的影响。结果从接骨木中分离得到了7个酚酸类化合物,分别鉴定为香草醛(vanillin,)、香草乙酮(aceto-vanillone,)、松柏醛(coniferyl aldehyde,)、丁香醛(syringaldehyde,)、对羟基苯甲酸(4-hydroxybenzoic acid,)、对羟基桂皮酸(4-hydroxycinnamic acid,)、原儿茶酸(protocatechuic acid,)。检测了化合物、~和对UMR106细胞活性的影响。结论7个酚酸类化合物均为首次从接骨木中分离得到,化合物可同时促进UMR106细胞的增殖和分化,化合物和可促进细胞的增殖,化合物和可促进细胞碱性磷酸酶活性。 Objective To study the antiostoporotic constituents in the stems of Sambucus williamsii and the effects of isolated compounds on proliferation and differentiation of the rat osteoblastic UMR106 cells. Methods Guided by the bioactivity on rat osteoblastic UMR106 cells, the constituents in the 60%o ethanol extract of the stems of S. williamsii were further separated with chromatographic methods. The structure of isolated compounds was identified by spectral analysis ; their effects on the proliferation and alkaline phosphatase (ALP) activities of UMR106 cells were observed. Results Seven phenolic acids and their derivatives were obtained, including vanillin ( Ⅰ ), acetovanillone ( Ⅱ ), coniferyl alcohol ( Ⅲ ), syringaldehyde ( Ⅳ ), 4-hydroxybenzoic acid ( Ⅴ ), 4-hydroxycinnamic acid ( Ⅵ ), protocatechuic acid ( Ⅶ ). The effects of compounds Ⅰ ,Ⅲ- Ⅴ , and Ⅶ on proliferation and ALP activities of UMR106 cells were determined. Conclusion These compounds are isolated from S. williamsii for the first time. Compound Ⅳ shows stimulating effects on both proliferation and ALP activities of UMR106 cells. Compounds Ⅰ and could only stimulate UMR106 cell prolifertion while compounds Ⅴ and Ⅶ could only induce ALP activities.
