摘要
目的检测糖皮质激素受体-α(GR-α)mRNA在甲状腺瘤和甲状腺正常组织中的定量表达,并探讨其在甲状腺瘤发生发展中的意义。方法我院住院手术治疗的22例甲状腺瘤患者。提取甲状腺瘤和甲状腺正常组RNA,应用实时荧光定量逆转录PCR(FQ-RT-PCR)检测GR-αmRNA定量表达。结果GR-α在甲状腺正常组织中(15.2±5.0)×105拷贝/μg比甲状腺瘤甲状组织中(2.4±4.9)×105拷贝/μg表达高,两组之间有明显差异(P<0.05)。结论正常甲状腺组织中存在糖皮质激素受体-αmRNA表达,生理量的糖皮质激素受体-α的基因效应减弱,可能是促进甲状腺瘤的发生发展的一个重要方面。
Objective To detect the expression of glucocorticoid receptor-α (GR-α) mRNA in thyroid adenomas and thyroid gland tissues and its role in the development of thyroid adenomas. Methods By using fluorescent quantitative PCR(FQ-RT-PCR), the expression of GR-α mRNA was detected in 22 previously diagnosed cases of thyroid adenomas and thyroid gland tissues. Results GR-α mRNA levels in the normal thyroid were significantly higher than in thyroid adenomas (P 〈 0.05). Conclusion GR-α was detectable in the thyroid adenoma and thyroid gland tissues. A downregulation of GR-α mRNA in thyroid adenoma seems to suggest that glucocorticoid is likely to play an important role in the developmerit of thyroid adenomas.
