苦参碱对肠癌HT-29细胞株增殖的抑制作用及其机制 被引量:24

摘要 目的观察苦参碱对肠癌HT-29细胞株增殖、凋亡的影响,探讨苦参抗肿瘤的作用机制。方法采用细胞培养、噻唑盐比色法(MTT)、流式细胞分析(FCM)、端粒酶重复片段扩增(TRAP)方法,分析测定苦参碱对肠癌HT-29细胞株增殖、凋亡及端粒酶活性的影响。结果用0、3、6、81、0μg/L苦参碱培养HT-29细胞24 h后,其增殖抑制率分别为:0、33.7%、59.0%、66.7%、69.0%。中效浓度(5μg/L)苦参碱培养HT-29细胞0、24、48、72 h后:细胞凋亡百分比分别是0.85%、15.31%、17.99%、26.58%;端粒酶活性随苦参碱处理时间延长而明显降低。结论苦参碱通过抑制DNA合成、诱导细胞凋亡、抑制端粒酶活性等多方面、多途径抑制肠癌HT-29细胞株增殖,达到抗肿瘤的作用。 Objective To study the anti-tumor mechanism of matrine by observing the effect of matrine on HT-29 cell line proliferation and apoptosis. Methods Using cells culture, MTT, FCM, TRAP methods, the effects of matrine on the proliferation, apoptosis and telomerase activity of HT-29 ceils were analyzed. Results After treatment for 24 h by matrine (0, 3, 6, 8, 10 μg/L), the inhibitory rate of proliferation was 0, 33.7%, 59.0%, 66.7%, 69% respectively. After the HT-29 cells were cultured with 5 μg/L matrine for 0, 24, 48, 72 h, the apoptosis rate was 0.85%, 15.31%, 17.99 %, 26.58 %, respectively. The telomerase activity was decreased with prolongation of matrine treatment time. Conclusion Matrine has the anti-tumor effect by suppressing HT-29 cells proliferation through inhibiting DNA synthesis, inducing apoptosis, inhibiting telomerase activity.
