腺病毒携带肝细胞生长因子基因不同途径转染对大鼠急性缺血心肌的影响 被引量:7

摘要 目的探讨腺病毒携带肝细胞生长因子基因(Ad-HGF)不同途径转染对大鼠缺血心肌的影响。方法将75只大鼠随机分为心肌内注射Ad-HGF组、注射携带绿色荧光蛋白(Ad-GFP)组、注射生理盐水空白对照组,心室腔注射Ad-HGF组、注射Ad-GFP组,各组分别于第3、7、142、1、28天各处死3只检查腺病毒的分布情况及进行血管计数。结果心肌内和心室腔注射Ad-HGF组第14、21、28天可见新生血管生成数量显著增加,与对照组相比差异有统计学意义(P<0.05)。心肌内注射Ad-GFP组术后3 d心脏绿色荧光最强,14 d消失,心腔内注射Ad-GFP组术后3 d心脏可见到范围较小的微弱绿色荧光,7 d消失。结论Ad-HGF可明显促进大鼠缺血心肌微小血管生成,心肌内直接注射途径仍优于心腔内给药。 Objective To investigate the effects of adenovirus-mediated hepatocyte growth factor (HGF) on acute ischemic myocardium in different pathways. Methods Seven-five rats were divided into 5 groups randomly. A volume of 100μl normal saline containing 1×10^9 plaque forming units (pfu) of recombinant Ad-HGF, adenovirus-mediated green fluorescence protein (Ad-GFP) or pure saline was injected directly into the ischemic myocardium, respectively. Ad-HGF and Ad-GFP was infused into left ventricular cavity, respectively. The transfected Ad-HGF expression was detected on the day 3, 7, 14, 21, 28 after operation. Results The fluorescent expression in Ad-GFP-injected group was stronger than that in Ad-GFP-infused group. The amount of capillary vessels was counted on the day 3,7, 14, 21,28 after operation. The number of vessels was more significantly increased in Ad-HGF-transfected group than that in control group. Conclusion The administration of Ad-HGF increased the number of vessels in regional myocardium. The transfection rate was lower in Ad-HGF infusion into left ventricular cavity group. The pathway of direct Ad-HGF injection was more effective on acute ischemic myocardium than that of Ad- HGF infusion into left ventricular cavity.
