Expression of Interleukin-10,-2 and TGF-β_1 in the Grafts of Rats during Acute Rejection of Heart Transplantation

摘要 目的研究白细胞介素2、10(IL-2、IL-10)与转化生长因子β1(TGF-β1)在大鼠心脏移植急性排斥反应中的表达.方法分两组建立同种大鼠颈部心脏移植模型:对照组(n=25)和新赛斯平组(CsA组,n=25),各5例观察移植心脏存活时间,于移植术后第1、5、7、14天分别取移植心组织各5例,半定量RT-PCR的方法动态检测移植心脏中IL-2、IL-10及TGF-β1的表达情况.结果对照组移植心存活时间为(8.6±1.5)d,CsA组移植心存活时间为(20.4±5.1)d,两组间移植心存活时间差异有显著性意义(P<0.01).两组中TGF-β1表达强度不同,对照组中表达较CsA组中弱,二者间差异有显著性意义(P<0.01).IL-2在对照组中先增高,于术后第5~7天达高峰,之后逐渐降至术后第14天的较低水平,而CsA组中IL-2则维持在较低水平.IL-10在对照组中表达维持于较低水平,CsA组中IL-10表达增高,于第7天达峰值,并维持高水平;两组间IL-10的表达差异有显著性意义(P<0.01).结论 IL-10和TGF-β1的产生和表达因CsA的干预而增加,IL-2的表达则受到抑制,CsA的干预明显延长了移植心脏的存活时间. Objective To study the expression of interleukin-10 (IL-10), IL-2 and transforming growth factor β1 (TGF-β1) in the grafts of the rats during acute rejection of heart transplantation. Methods Allografts were divided into 2 groups: control group (n=25) and cyclosporin A-treated group (n=25). Hearts from SD rats were transplanted into cervical sites in Wistar recipients and the survival time of the allografts was observed. The local expression of TGF-β1 , IL-2 and IL-10 was detected at day 1, 5, 7, 14 by reverse transcription polymerase chain reaction (RT-PCR) respectively. Results The survival time of the allografts was (8.6±1.5) days and (20.4±5. 1) days in control group and cyclosporin A-treated group respectively (P〈0.01). The levels of IL-10 and TGF-β1 expression were up-regulated in the cyclosporin A-treated group vs control group (P〈0.01). The expression of IL-2 was down-regulated in cyclosporin A-treated group vs control group (P〈0.01). Conclusion The expression and production of TGF-β1 and IL-10 are up-regulated with the administration of cyclosporin A, while IL-2 down-regulated, which are favorite to the heart graft survival.
