葛根药用成分异黄酮的HPLC指纹图谱分析 被引量:7

摘要 采用高效液相色谱法对葛属植物野葛和粉葛块根的主要药用成分异黄酮化合物的指纹图谱进行了研究。结果表明,以乙腈-0.5%乙酸为流动相,采用梯度洗脱的方法,葛根异黄酮化合物的各种组分可得到良好的分离;通过对10批次野葛药材HPLC图谱的检测,共确定了15个色谱峰作为野葛的共有指纹峰,根据中药指纹图谱技术的基本原则和方法,建立了野葛的HPLC指纹图谱;与不同产地野葛的HPLC图谱的比较,供试样品的指纹图谱基本一致;而与粉葛样品的HPLC图谱存在较大的差异;表明得到的色谱图可以作为野葛药材专属性的指纹图谱,为葛根的药材鉴别和质量控制提供了实验依据。 The main chemical constituents, i.e. isoflavonoids of a traditional Chinese medicine, kudzu, were analyzed by HPLC-FPC. The results showed that the components of Pueraria lobata (Willd.) Ohwi. were well separated with the eluted gradient ofacetonitrile - 0.5 % acetic acid. On the basis of HPLC-FPC results of P. lobata (ten batches) from Anhui Province of P. R. China, and the basic requirements of criteria and methods for fingerprint .chromatography, fifteen peaks were selected as the common fingerprint peaks, and the HPLC-FPC of P. Iobm, was determined. Comparison of the HPLC-FPC between P. lobata and other kudzu cultivars (or varieties) from different sources showed that those peaks existed in all the samples. The significant differences of retention time and area percentage of common fingerprint peaks between P. lobata and P. thomsonii were also investigated, from which the two species could be distinguished effectively. Thus, the HPLC-FPC can be used for identification and quality assessment of P. Iobata.
