黄体酮与维生素K_3对家兔输尿管动作电位和尿流量的影响 被引量:1

Effects of progesterone and vitamin K_3 on ureteral action potential and urinary flow in rabbits

摘要 目的:以兔输尿管平滑肌为观察对象,探讨黄体酮与维生素K3对其电活动及尿流量的影响。方法:实验于2004-03/10在郧阳医学院机能实验中心实验室完成。选择健康家兔29只,随机分为4组,即生理盐水对照组5只;黄体酮组8只;维生素K3组8只;黄体酮+维生素K3组8只。4组均先记录10min正常值作为对照值,然后生理盐水对照组给予生理盐水2mL/kg,耳缘静脉注射;黄体酮组予黄体酮20mg静脉注射;维生素K3组予维生素K38mg/kg静脉注射;黄体酮+维生素K3组,分别给予黄体酮20mg、维生素K38mg/kg静脉注射。采用在体电活动记录给药后1,5,10,15,30,45,60及90min输尿管平滑肌动作电位变化及尿流量变化。结果:最终进入统计分析的家兔保持为4组共29只,无缺失值。黄体酮组在第10,15,30,45,60及90分钟输尿管电活动的频率与给药前比较显著较低(P<0.05),60及90min的尿流量与给药前比较显著增加(P<0.05)。维生素K3组在第15,30,45,60及90分钟输尿管电活动的频率与给药前比较显著较低(P<0.05),在60及90min的尿流量与给药前比较显著增加(P<0.05)。黄体酮与维生素K3合用能显著降低输尿管电活动的频率,其中5,10,15,30,45,60及90min与给药前比较差异显著(P<0.05),并且在15,30,45,60及90min的尿流量与给药前比较明显增加(P<0.05)。结论:黄体酮与维生素K3对兔输尿管平滑肌有明显的舒张作用,可使输尿管动作电位频率降低,尿流量增加,而两药合用则对舒张输尿管平滑肌与尿流量增加起协同作用。 AIM:To study the effects of progesterone and vitamin K3 on ureteral action potential and urine flow,taking smooth muscle of ureter in rabbits as observation objects. METHODS:The experiment was conducted at the central laboratory for functional experiment of Yunyang Medical College from March to October 2004.Twenty-nine healthy rabbits were selected and assigned randomly into 4 groups: saline control group with 5 rabbits, progesterone group with 8 rabbits, vitamin Ks group with 8 rabbits and progesterone+vitamin Ks group with 8 rabbits. Normal value for 10 minutes was recorded in the four groups as control value,and then the rabbits in the saline control group were injected with 2 mL/kg saline into a marginal ear vein. Those in the progesterone group were injected with 20 mg progesterone into vein. Those in the vitamin K3 group were injected with 3 mg/kg vitamin K3 into vein. Those in the progesterone+vitamin K3 group were injected with 20 nag progesterone and 8 mg/kg vitamin K3 into vein, respectively. The changes of action potential of smooth muscle and urinary flow were recored with electrical activity after administration for 1, 5, 10, 15, 30, 45, 60 and 90 minutes. RESULTS:Totally 29 rabbits from four groups were involved in the statistical analysis,without deletion. the progesterone group at the 10^th, Frequency of ureteral action potential in 15^th, 30^th, 45^th, 60^th and 90^th was lower significantly than that before administration (P〈0.05). The urinary flow at the 60^th and 90^th minutes decreased significantly as compared with that before administration (〈0.05).Frequency of ureteral action potential in the vitamin K3 group at the 15^th,30^th, 45^th, 60^th and 90^th minutes was lower than that before administration (P〈0.05), and the urinary flow at the 60^th and 90^th minutes increased significantly than before administration (P〈0.05). Combination of progesterone and vitamin K3 could reduce significantly the frequency of ureteral action potential, in which at the 5^th, 10^th, 15^th, 30^th, 45^th, 60^th and 90^th minutes the difference was significant as compared with preadministration (P〈0.05 ). Besides, the urinary flow at the 15^th, 30^th, 45^th,60^th and 90^th minutes increased significantly as compared with pre-administration (P〈0.05). CONCLUSION:The progesterone and vitamin K3 have obvious diastole action on rabbit smooth muscle of the ureter,decrease action potential of ureter, and increase the urinary flow. The combination of the two drugs can have synergistic aetion on the increase of diastole ureter of smooth muscle and urinary flow.
