三疣梭子蟹养殖群体同工酶的组织特异性及生化遗传分析 被引量:11

摘要 应用垂直平板聚丙烯酰胺凝胶电泳技术(PAGE),研究分析了取自浙江舟山朱 家尖养殖区的三疣梭子蟹5种组织中10种同工酶以及肌浆蛋白的表达情况,并进行 生化遗传分析.结果显示,在对40只梭子蟹所检测出的10种同工酶的25个基因座 位中,ADH、ACP-1、CAT-2、EST-2、FDH-2和SOD共6个基因座位为多态,其多态座 位比例(P)为24%,平均杂合度期望值(H)为0.106 6.同时在CAT、LDH、ACP、FDH 4种酶及肌浆蛋白(Pm)中表现出显著的组织特异性. Portunus trituberculatu samples were collected from the cultural net in zhoushan, Zhe-jiang Province. Polyarylamide gel electrophoresis was used to detect the expression of isozymes in 5 tissues of 40 crabs : gill, liver , heart, hemocytoblast , muscle. 10 isozymes ( ADH, ACP, CAT, ATP, EST, FDH, GDH, LDH, MDH, SOD) and Pm were analyzed and the biochemical genetic results showed that 10 isozymes were coded by 25 gene loci ,6 of which (ADH , ACP-1, CAT-2, EST-2, FDH-2, SOD) were found polymorphic. The mean proportion of polymorphic loci was 24 % , and the average heterozygosity per locus was 0. 106 6. Significant tissue specificity was observed in 4 isozymes ( CAT, LDH, ACP, FDH ) and Pro.
