摘要
目的:对复方白细胞介素-2(IL-2)长循环脂质体的制备方法及其质量控制进行研究。方法:用氯仿注入合并硫酸铵梯度法制备复方白细胞介素-2长循环脂质体。用紫外-Sephadex法测定脂质体中猪苓多糖的含量和包封率;用酶联免疫法(ELISA)测定IL-2的含量和包封率。结果:长循环脂质体中猪苓多糖和IL-2的含量分别为4.02g.L-1和1 215IU.mL-1;猪苓多糖和IL-2的包封率分别为35.5%和33.2%。结论:用氯仿注入合并硫酸铵梯度法可制得包封率高的脂质体。紫外-Sephadex法可用于测定猪苓多糖的含量和包封率,酶联免疫法可用于测定IL-2的含量和包封率;方法简便易行,准确度高,重现性好。
OBJECTIVE To prepare the composite IL-2 long circulating liposome (LCL) and to study its quality control. METH- OIlS The LCL was prepared by means of chloroform injection combined with the ammonium sulphate gradients. The content and encapsulation efficiency of Polyporus umbellate polysaccharides were determined by sephadex coloumn-ultraviolet method. The content and encapsulation efficiency of IL-2 were determined by ELISA method. RESULTS The content of Polyporus umbellate polysaccharides and IL-2 in LCL was 4. 02 g·L^-1,1 215 IU·mL^-1 , respectively. The encapsulation efficiency of Polyporus umbellate polysaccharides and IL-2 was 35.5%, 33.2%, respectively. CONCLUSION The LCL with high entrapping efficiency can be prepared by means of chloroform injection combined with the ammonium sulphate gradients. The ELISA method is suitable for the quality control of IL-2 and the sephadex column-ultraviolet method is suitable for the quality control of Polyporus umbellate polysaccharides.
